PE-22-28 is a synthetic analogue of spadin, a peptide derived from the sortilin propeptide. Research attention centres on TREK-1 potassium channel blockade, a mechanism proposed as a faster-acting antidepressant pathway than monoamine reuptake inhibition, with rodent behavioural work reporting effects within days rather than weeks.
